Leptospirosis là một bệnh truyền nhiễm chung của nhiều loài động vật và người, hiện diện phổ biến khắp thế giới. Sự lưu hành huyết thanh học của Leptospira được ghi nhận trên chó nuôi ở khu vực đồng bằng sông Cửu Long (ĐBSCL). Gần đây, xuất hiện những trường hợp chó nghi mắc leptospirosis với triệu chứng vàng da, xuất huyết niêm mạc, chảy nước mắt, đau cơ và rối loạn thận-niệu… đã được đưa đến khám và điều trị tại các bệnh xá thú y trong khu vực. Mục tiêu của nghiên cứu này là phát hiện xoắn khuẩn Leptospira gây bệnh trên chó. Một trăm mười một (111) mẫu nước tiểu chó có hiệu giá MAT≥1400 đã được thu thập để phát hiện sự hiện diện của xoắn khuẩn bằng phương pháp quan sát dưới kính hiển vi nền đen. Xoắn khuẩn ở những mẫu nước tiểu dương tính được nuôi cấy tiếp trên môi trường EMJH, giám định xoắn khuẩn bằng PCR và giải trình tự gen 16S rRNA để định danh loài. Kết quả quan sát bằng kính hiển vi nền đen đã phát hiện xoắn khuẩn hiện diện trong 63/111 (56,75%) mẫu nước tiểu, nhưng chỉ nuôi cấy Leptospira thành công trên môi trường EMJH từ 3 trong số 63 mẫu nước tiểu này (chiếm tỷ lệ 4,76%). Kết quả giám định phân tử xoắn khuẩn đã phát hiện Leptospira interrogans trong hai mẫu nước tiểu của chó có biểu hiện nhiễm bệnh lâm sàng và Leptospira fainei hiện diện trong một mẫu nước tiểu của chó chưa biểu hiện bệnh, trong đó L. interrogans thuộc nhóm xoắn khuẩn gây bệnh trực tiếp và L. fainei thuộc nhóm xoắn khuẩn gây bệnh cơ hội. Đây là kết quả phân lập và định danh xoắn khuẩn Leptospira gây bệnh từ nước tiểu chó đầu tiên ở Việt Nam.

Leptospirosis is a popularly zoonotic disease of the worldwide. The sero-prevalence of leptospirosis was reported in the domestic dogs in the Mekong Delta. Recently, the dogs suspecting leptospirosis with the clinical syndromes, such as jaundice, mucosal hemorrhage, oculo-nasal discharge, abdominal pain and renal dysfunction were recorded in the veterinary clinics in the investigating area. The objective of this study was to detect the Leptospira species causing leptospirosis in the dogs. One hundred and eleven (111) urine samples were collected from the suspected dogs to detect the presence of leptospira by a dark-field microscopy; leptospira from the positive samples was inoculated in the EMJH medium, then Leptospira species were identified by PCR assay, and 16S rRNA gene sequence. The results of dark-field microscopic examination showed that leptospira was present in 63 out of 111 (56.75%) urine samples, however leptospira was cultured successfully on EMJH medium from only 3 out of 63 positive urine samples, accounting for 4.76%. The results of analysing leptospira gene sequences revealed two species, i.e. Leptospira interrogans and Leptospira fainei presenting in the urine of the investigated dogs, of which L. interrogans belonged to the mainly pathogenic group and L. fainei belonged to the associated pathogenic group. This is the first successful study on isolation and identification of pathogenic Leptospira from the urine of the dogs suspecting leptospirosis in Viet Nam.
