Phân tích nồng độ beta-hCG có ý nghĩa quan trọng trong chẩn đoán và sàng lọc sơ sinh. Trong nghiên cứu này chúng tôi tối ưu hóa các điều kiện đính kháng thể hCG lên đế polystyrene (PS) bằng phương pháp hấp phụ để sử dụng phân tích beta-hCG bằng kỹ thuật miễn dịch huỳnh quang. Sử dụng kết hợp kháng thể hCG đánh dấu với Eu(III) và dung dịch tăng cường tín hiệu huỳnh quang chúng tôi đã lựa chọn được điều kiện hấp phụ tối ưu là nồng độ dung dịch là 2,5μg/L, thời gian hấp phụ là 24 giờ. Hoạt tính của kháng thể hCG cố định trên đế PS thay đổi không đáng kể khi lưu giữ ở 4oC trong 3 tuần. Quy trình phân tích sử dụng đế PS cố định hCG có độ chọn lọc đặc hiệu với beta-hCG, có độ lệch trung bình -2,1% so với Kit phân tích chuẩn, có giới hạn phát hiện (LOD) và giới hạn phân tích (LOQ) lần lượt là 11,9 và 17,9 ng/ml. Kết quả này cho phép sử dụng quy trình phân tích tự xây dựng dựa trên đế PS cố định hCG để phân tích beta-hCG với độ chính xác cao, chi phí thấp với công nghệ hoàn toàn chủ động.

Measurements of beta form of human chorionic gonadotropin (hCG) provide important information in a variety of clinical situations and prenatal screening. In this study, we optimized conditions to absorb hCG antibody onto polystyrene (PS) substrates for immunofluorescence analysis of hCG. The conditions were monitored by evaluating the photoluminescent (PL) intensity obtained upon using Eu(III) labelled hCG antibody and PL enhancement solution. The optimized concentration of hCG antibody in the absorbent solution was found to be 2.5μg/L and the absorption time was 24 hours. The reactivity of the absorbed hCG antibody decreased negligibly after three weeks of storing at 4oC. The immunofluorescence procedure based on the hCG/PS substrates was highly selective to beta-hCG, giving good analytical results when compared with standard Kit. Lower limit of detection (LOD) and lower limit of quantification (LOQ) of the analytical procedure were estimated to be 11.9 and 17.9 ng/ml, respectively. The results demonstrated in this paper provide a cost-effective and precise method for quantification of beta-hCG based on hCG antibody absorbed on PS substrates.
