My Lam, Tuyen Quang hot spring is located in the stone mountain in Tuyen Quang province. The temperature and pH of water from its well are range from 65 to 67°C and from 8.0 to 8.2, respectively. The mineral content of water is high and diversed. From water and mud samples of My Lam hot spring, the thermophilic bacteria were enriched on LB and extremely medium for two weeks at 65oC. Ten thermophilic bacterial strains producing cellulase and amylase (denoted ML01 to ML10) was isolated and purified. Among ten purified strains, three strains (ML03, ML05 and ML07) were determined to produce the highest cellulase and amylase activity. The observed morphological, physiological and biochemical characteristics indicated that the ML03, ML05 and ML07 strains belong to Geobacillus genus. However, to be able to classify these trains to species, the study of 16s rRNA gene sequence has been conducted. The result showed that the sequence of ML03, ML05 strain have 99 percent homology with Geobacillus caldoxylosilytjcus species and named as G. caldoxylosilytjcus ML03 and G.caldoxylosilyticus ML05. whereas, the 16s rRNA gene sequence of ML07 strain have only 97 percent similarity with species of Geobacillus genus and was called Geobacillus sp. ML07. Simultaneously, the 16s rRNA gene sequences of ML03, ML05 and ML07 have been submitted on gene bank (NCBI) with accession numbers are Fj389674, Fj389676 and Fj389676, respectively.
