Nghiên cứu chuyển gen tạo rễ tơ cây xáo tam phân (Paramignya trimera) thông qua Agrobacterium rhizogenes K599.

Nghiên cứu được tiến hành nhằm cảm ứng tạo rễ tơ cây xáo tam phân (Paramignya trimera) nhờ vi khuẩn Agrobacterium rhizogenes K599 và khảo sát một số yếu tố ảnh hưởng đến sự tăng sinh khối rễ tơ. Ba loại vật liệu khác nhau là mô sẹo, lá mầm và rễ cây con in vivo được sử dụng làm nguồn lây nhiễm cảm ứng tạo rễ tơ. Kết quả cho thấy, rễ cây con in vivo là loại vật liệu thích hợp để cảm ứng tạo rễ tơ cây xáo tam phân. Mật độ vi khuẩn cho tỷ lệ mô rễ cảm ứng tạo rễ cao nhất (42,7%) tương ứng với giá trị mật độ quang OD600=0,6 trong thời gian lây nhiễm là 30 phút. Các dòng rễ tơ có khả năng tăng trưởng nhanh, ổn định và hiệu quả hình thành rễ tơ cao nhất khi nuôi cấy trong môi trường WPM/2 không bổ sung chất điều tiết sinh trưởng ở điều kiện tối trong 6 ngày. Chiếu sáng bằng đèn huỳnh quang ức chế sự hình thành rễ tơ giai đoạn ủ cảm ứng nhưng thúc đẩy quá trình này khi áp dụng ở bước loại bỏ vi khuẩn. Các dòng rễ tơ chuyển gen đã được kiểm chứng nhờ kỹ thuật PCR với cặp mồi chuyên rolC.
