Leptospirosis is an emerging zoonosis disease. The definitive diagnosis of Leptospirosis has traditionally depended upon the isolation of Leptospires from clinical specimens or detennination of antibodies against Leptospira in serum samples. These approaches require expertise not routinely available in clinical laboratories and usually result in delayed diagnosis. In order to overcome these limitations, a PCR assay was developed using a 423 bp target on the LipL32gene, which is encoded for lipoprotein LipL32 and conserved among pathogenic serovars of Leptospira. Fifteen pathogenic and 1 non-pathogenic Leptospiral serovars and some other bacteria strains were tested. The assay was specific for Leptospira and may discriminate pathogenic and non-pathogenic species. Applied this assay to detect Leptospiral DNA from pig kidney specimens collected from slaughter-house in some provinces in the South Central region and Highland of Vietnam shown that there were 14.43 percent of specimens to be positive.
