Mahonia bealei, ITS (Internal Transcribed Spacer), conservation, Berberidaceae

Phân tích đặc điểm phân tử vùng gen ITS-rDNA của loài Hoàng liên ô rô lá dày. Kết quả cho thấy vùng gen ITS dài 700 bp có chứa 24,1% Timin, 28,4% Guamin, 23,4% Adenin và 24,1% Cistezin, là vùng gen phù hợp để sử dụng trong công tác xác định loài và nghiên cứu mối quan hệ di truyền cho các loài họ Berberidaceae. Sự khác biệt di truyền trung bình giữa chi Mahonia và chi Berberis là 2,3%, với các chi khác là 17% đến 22%. Cặp mồi đặc hiệu khuếch đại vùng gen ITS dài 300 bp loài Mahonia bealei đã được phát triển và tổng hợp. Trình tự vùng gen ITS-300 của Hoàng liên ô rô lá dày đã được đăng ký lên ngân hàng gen thế giới với mã số MT 008067.

 In this research, we study on molecular characteristics of ITS-rDNA region of Mahonia bealei. The gotten result showed that, the ITS with 700 bp in length contained 24.1% Timin, 28.4% Guamin, 23.4% Adenin and 24.1% Cistezin. The average genetic difference between Mahonia genus and Berberis genus is 2,3% while with other genera is from 17% to 22%. The gotten result also demonstrated this ITS region can be useful and suitable for identification and study phylogenetic of Berberidaceae. Specific primer to amplify the ITS genome in the length of 300 bp of Mahonia bealei species aslo have been developed and synthesized. This ITS-300 genome sequence of Mahonia bealei was registered on the Gene Bank with accession number as MT 008067.
