In this study, poly(ethylene glycol) (PEG) was activated by p-nitrophenyl chloroformate (NPC) and then partially substituted by tyramine (TA). The PEG derivative was grafted onto chitosan via NPC-activated residue of PEG. The grafted method could gain up to 10 PEG-TA moieties per 100 glucosamine units. Poly(ethylene glycol)-chitosan hydrogel was prepared in situ via an enzymatic reaction of tyramine moieties on the grafted polymer in the presence of horseradish peroxidase (HRP)/hydrogen peroxide (H2O2). The in situ gel formation was occurred within a time period of 10 s to 32 s matching with amount of the grafted TEG-TA. For in vitro study, the hydrogels showed a good cytocompatibility. These hydrogels could be promising injectable biomaterials in biomedical applications due to its adjustable gelation times and high biocompatibility.
