Hệ chuyển giao thuốc dạng màng phim tan nhanh trong miệng ngày càng được quan tâm nghiên cứu để sử dụng trên bệnh nhân khó nuốt như người già, trẻ em. Nghiên cứu này được thực hiện nhằm xây dựng công thức điều chế màng phim tan nhanh trong miệng chứa clorpheniramin maleat 4 mg. Phương pháp nghiên cứu: Màng phim có các thành phần chính gồm polyme, chất hóa dẻo và dược chất được điều chế bằng phương pháp bay hơi dung môi. Ảnh hưởng của thành phần công thức tới tính chất màng phim được khảo sát bằng thiết kế thực nghiệm. Trạng thái vật lý hoạt chất được xác định bằng phân tích nhiễu xạ tia X, nhiệt quét vi sai và quang phổ hồng ngoại biến đổi Fourier. Quá trình hình thành màng phim được xác định nhờ kính hiển vi điện tử quét. Kết quả: Nồng độ polyme, loại polyme có ảnh hưởng đến thời gian tan của màng. Độ bền kéo của màng phim bị tác động b i loại polyme và loại chất hóa dẻo. Độ giải phóng hoạt chất không bị ảnh hư ng b i thành phần công thức. Công thức điều chế đã được xác định gồm HPMC E15 (0,83%), PVA (1,67%), clorpheniramin maleat (0,41%), propylen glycol (0,25%), aspartam (0,56%), acid citric (0,04%) và nước cất vừa đủ để tạo ra màng phim có thời gian tan trung bình xấp xỉ 7 giây, độ bền kéo 26,67 N/mm2 và phóng thích hơn 90% dược chất sau 5 phút. Màng phim có cấu trúc xốp và dược chất tồn tại dạng phân tử trong dung dịch rắn với mạng polyme và có cấu trúc hóa học không đổi. Kết luận: Công thức phù hợp để điều chế màng phim tan nhanh trong miệng chứa clorpheniramin maleat 4 mg đã được xác định. Quá trình hình thành và trạng thái vật lý của hoạt chất trong màng phim đã được khảo sát.

Oral fast dissolving film (ODF) delivery system is increasingly interested in using for patients who have dysphagia such as pediatrics and geriatrics. This study aims to develop a formulation of oral fast dissolving film containing chlorpheniramine maleate 4 mg. Materials and methods: Oral fast dissolving thin films contained main components such as polymer, plasticizer and chlorpheniramine maleat which were produced by using solvent casting technique. Formula and factors affecting the film’s properties were established by Design Expert v 2. software. The physical state of active ingredient was determined by X-ray diffraction, differential scanning calorimetry and Fourier transforms infrared spectroscopy. The film formation mechanism was determined by scanning electron microscope. Results and discussion: The polymer’s type and its concentration affected the film’s dissolving time. The film tensile strength depended on the type of polymer and plasticizer. Besides, the film dissolution was not affected by any factors. The formulation of chlorpheniramine maleate loaded film was se-lected based on the setting conditions. It includes HPMC E15 (0.83%), PVA (1.67%), chlorpheniramine maleate (0.41.), propylene glycol (0.25%), aspartame (0.56%), acid citric (0.04%), distilled water (96.24%). As a result, the film had average dissolving time of 7 seconds approximately, tensile strength of 26.67 N/mm2 and the drug substance releases more than 90% after 5 minutes. The film had a porous structure. The active substance remained molecule of solidsolution in the polymer lattice with invariable chemical structure. Conclusion: The study established the formulation of oral fast dissolving film which contain chlorpheniramine maleate 4 mg. Additionally, the film formation mechanism and the physical state of the active ingredient in the film were clearly clarified.
