Quy trình duplex PCR xác định nhiễm trùng Mycoplasma hyopneumoniae - Mycoplasma hyorhinis trên mẫu phổi, dịch khớp và dịch xoang miệng trên heo

Nghiên cứu thực hiện nhằm xây dựng và tối ưu hóa quy trình duplex PCR phát hiện đồng thời Mycoplasma hyopneumoniae (Mhp) và Mycoplasma hyorhinis (Mhr) trên heo. Duplex PCR được xây dựng thành công với các thông số tối ưu như sau Nồng độ mồi Mhp16s-F, Mhp16s-R, Mhr37p-F, và Mhr37p-R lần lượt là 0,5 μM; 0,5 μM; 0,5 μM; và 0,5 μM. Chu trình nhiệt tối ưu cho phản ứng d-PCR bao gồm giai đoạn tiền biến tính ở 94 oC trong 3phút, biến tính ở 94 oC trong 1 phút, bắt cặp ở 45 oC trong 1 phút, kéo dài ở 72 oC trong 1 phút và giai đoạn hậu kéo dài ở 72 oC trong 5 phút, lặp lại trong 35 chu kỳ. Quy trình d-PCR có đặc hiệu và độ nhạy cao với khả năng phát hiện tối thiểu 20,9 copies/ng (Mhp) và 15,0 copies/ng (Mhr). Quy trình d-PCR có tính ứng dụng cao khi 11/11 mẫu thực địa cho kết quả đồng nhất với quy trình s-PCR.
