Southern rice black-striked dwarf virus-SRBSDV was a new virus species of genus Fijivirus in the Reovidae family which caused serious rice losses in an area of millions of hectares throughout southern China since 2001 and first got outbreak in northern, central Vietnam in 2009-2010 with an area of tens of thousands of hectares. At present, there is no specific antibodies against to SRBSDV in the form of trade. To develop diagnostic techniques by ELISA, in a previous study, optimal conditions for expression and purification of gene p10 encoding outer capsid protein of SRBSDV were realized. In this study, recombinant outer capsid protein of SRBSDV was used as antigen to inject into 6-8 week old mice. The specific antibodies IgG against to SRBSDV generates in serum was collected by mice blood centrifuge after 4 weeks injected recombinant proteins and purified by column chromatography over protein A-Sepharose. The titer of specific antibodies IgG against to SRBSDV were checked by Western blot and Dot-blot as it can recognizes recombinant P10 protein with dilution 1: 6000. The results of study opens up a new opportunity for the diagnosis SRBSDV by ELISA.
