Species identification and genetic diversity analysis play an important role in disease control. Twenty Colletotrichum spp. isolated from various grape varieties are used for species identification by specific PCR using ITS4/Cglnt primers to amplify a DNA fragment of 450 bp in the Internal Transcribed Spacer (ITS) region and genetic diversity analysis by RAPD (Random Amplified Polymorphic DNA) marker using seven random primers. Identification result by specific PCR reaction shows that all the twenty Colletotrichum spp. isolates using in this study are C. gloeosporioides. Study on genetic diversity of C. gloeosporioides using RAPD marker has obtained a total of 764 bands, in which 56 bands have different sizes. In 56 bands of different sizes, there are 44 polymorphic bands accounting 78.57 percent and 12 simplex bands accounting 21.42 percent. Genetic similarity coefficients fluctuates from 0.41-0.8, in which a total of 190 pairs consists of 7 pairs with genetic similarity coefficients of 0.4 accounting 3.68 percent, 61 pairs with genetic similarity coefficients of 0.5 accounting 32.11 percent, 82 pairs with genetic similarity coefficients of 0.6 accounting 43.16 percent, 38 pairs with genetic similarity coefficients of 0.7 accounting 20.00 percent and 2 pairs with genetic similarity coefficients of 0.80 accounting 1.05 percent. Genetic relation analysis of 20 Colletotrichum spp. isolates in this study is split into 3 separated groups: Group 1 consists of 2 species, group 2 consists of 12 spp., group 3 consists of 5 spp., and a isolate is outside of genetic tree.
