To establish a procedure for diagnosis of prostate cancer by PCA3 score we used cell lines: LNCaP, Hela, HT29, NB4, HL-60, K562, THP-1 and urine samples from 30 subjects. The results showed that gene PCA3 was expressed only in LNCaP cells but others. Minimum level of LNCaP could be detected gene PCA3 was 10 exponent 3 cells/ml. mRNA PCA3 level in prostate cancer patients was significant higher compared to healthy control and benign prostatic hyperplasia (1436.5 + or - 1852.7 vs 15.3 + or - 30.8 and 54.5 + or - 90.34; p0.05). AUC of PSA is higher compared to AUC of PCA3 Score, however, PCA3 score is more sensitivety.
