This study was conducted to develop a process of Gerbera breeding by tissue culture methods. The results of this study show that the best sterilization methods is to neutralize Javen water with the concentrations of 20 percent in 30 minutes with the higtlest alive sample rate of 43,33 percent. The best medium for the shoot is to add 0.5 mg/l BA with the result number of 6.53 shoots and the height of 5.11cm after 42 days of culture and the status of big buds with dark green leaves. The best medium for root development is to add 0.3 to 0.5 mg/l NAA with the highest number of roots and root length. Acclimatization obtains the best result on 100 percent coconut fiber after 42-day with the survival rate of 85.71 percent.
