Trong nghiên cứu này, chủng chủ Bacillus subtilis và gen chỉ thị celB được sử dụng để đánh giá ảnh hưởng của số lượng Histidine trong đuôi dung hợp ở đầu C lên sự biểu hiện tiết Endoglucanase B. Kết quả nghiên cứu cho thấy chủng B. subtilis 1012 phù hợp cho sự biểu hiện CelB hơn khi so với chủng B. subtilis WB800N. Đuôi 6His có ảnh hưởng làm giảm sự biểu hiện tiết CelB nhiều hơn hai đuôi 5His và 4His. Ngược lại, CelB-6His cho khả năng bám hạt Ni-NTA tốt hơn, ít thất thoát nhất (4,78%) trong khi CelB-4His lại có sự rò rỉ khi tinh chế bằng hạt Ni-NTA ở mức cao nhất (16,1%).

In this research, the host cell Bacillus subtilis and the reporter gene celB were used for evaluating the impact of the amount of histidine in C-terminal fusion tag on the secretion of Endogucanase B. The results show that B subtilis 1012 is more appropriate for secreting CelB than B. subtilis WB800N. Among three kinds of His tag (4His, 5His, 6His), 6His tag has the most impacts on decreasing the secretion of CelB. In contrast, the surveying of binding capacity of these proteins demonstrates that CelB-6His has the best binding capacity and the fewest leakage (4.78%) while CelB-4His has the most leakage (16.1%) in purifying process by Ni-NTA.
