Thẩm định quy trình xác định đồng thời Zidovudin và Lamivudin trên hệ chất mang nano dendrimer G3.0-F127@AZT/3TC bằng phương pháp HPLC-PDA

Nghiên cứu được thực hiện nhằm xác định đồng thời Zidovudin (AZT) và Lamivudin (3TC) trên nền dendrimer PAMAM biến tính với pluronic F127, ứng dụng làm thuốc chống HIV có kiểm soát. Một phương pháp xác định AZT và 3TC bằng kỹ thuật HPLC-PDA nhanh, chính xác với độ tin cậy và độ chọn lọc cao đã được xây dựng và thẩm định theo hướng dẫn của ICH (Hội nghị quốc tế về hài hòa các thủ tục đăng ký dược phẩm sử dụng cho con người). Quá trình sắc ký được thực hiện trên cột pha đảo C18 sử dụng pha động là dung dịch đệm phosphat pH 2,5 - methanol (6535), tốc độ dòng 1,0 ml/phút. Bước sóng phát hiện 266 nm. Thời gian lưu của AZT và 3TC lần lượt là 3,5 và 4,3 phút với độ phân giải 3,15. Đường tuyến tính từ 2,16 đến 324,53 μg/ml đối với AZT, và 2,54 đến 152,10 μg/ml đối với 3TC (r=1,000); tỷ lệ phục hồi trung bình là 100,30% (RSD=0,36%) đối với AZT và 3TC là 99,73% (RSD=0,50%). Kết quả thẩm định cho thấy đạt độ đúng, chính xác và tính phù hợp của phương pháp trong phân tích hoạt chất AZT và 3TC trên hệ chất mang nano dendrimer PAMAM G3.0-F127@AZT/3TC là đạt yêu cầu.
