Background: Prostate Cancer Gene 3 (PCA3) has been used for diagnosis of prostate cancer, howevere, there isn't any lab in Vietnam perfomed this experiment. Aims: To establish the procedure for applying PCA3 in diagnosis of prostate cancer. Subiects and methods: LNCaP cell, 10 healthy control; 10 Benign prostatic hyperplasia (BPH) and 10 patients with Prostate Cancer. LNCaP cells were culture and diluted into women urine. mRNA PCA3 and PSA were measured by Real Time PCR. PCA3 Score = mRNA, PCA3:mRNA PSA. Result: Minimum number of LNCaP cells could be detected mRNA PCA3 and PSA were 103 celVml and 102 celVml, respectively. PCA3 Score in Prostate Cancer group was significant higher comparison to healthy control and BPH (383,8 + or - 457.5 vs 7.7 + or - 7.4 and 10.9 + or - 12.7; p0,05).
